I think there's one thing might be a problem here. In your Experience story you said you injected the mouse zygote with the NLS-NgAgo plasmid from addgene with the guide DNA...
I bet it cannot be reproduced too. However, your experiment is inconclusive for it. Oocyte cannot recapture the cell lines. You can only say NgAgo might not work for oocyte based on your results...

Did't work in Yeast either. I will test again with codon optimized NgAgo for Yeast. Will post the results soon
I think I have an idea why this paper is irreproducible. This simply doesn't work. I've done some experiments to try reproduce the findings in Han's paper...
